科研领域描述
淋巴细胞免疫受体(包括抗体和T细胞受体)的产生依赖于RAG内切酶起始的V(D)J重排过程。在V(D)J重排过程中,RAG会主动切开DNA双链,而这些DNA双链断裂的修复依赖于体内的DNA修复途径。一旦这些DNA损伤不能正确修复,则可能形成染色体易位,从而高调癌症基因,如C-MYC等,进而导致淋巴瘤等癌症的发生。为了研究淋巴细胞内RAG介导的程序性损伤的调控机制及其相关的肿瘤发生机理,我们开发了基于二代高通量测序的染色体易位/DNA双链断裂测序方法,计划用该方法来高灵敏度地追踪DNA双链断裂在细胞内的发生、修复过程并研究其促癌机理。此外,我们计划利用基于单细胞测序的DNA复制和细胞转录测序技术以研究DNA复制和细胞转录与淋巴细胞基因组稳定性的关系,并进一步阐明基因组三维结构在RAG活性、DNA复制和细胞转录方面的关键调控机制。V(D)J重排的相关研究有助于了解免疫受体的发育和特异性产生的分子机制,我们开发了可定量的免疫组化库测序新技术并计划利用该方法深入研究淋巴细胞免疫受体发育机制以及实现针对特定抗原的特异性免疫受体的筛选和应用。
